Overview

Human papillomavirus type 16 L1 virus-like particle vaccine is a prophylactic biologic vaccine designed to prevent infection with HPV type 16, which is associated with approximately 50% of cervical cancers and high-grade cervical intraepithelial neoplasias. The vaccine consists of virus-like particles (VLPs) composed of the L1 major capsid protein of HPV-16, which are non-infectious particles that resemble HPV virions. These VLPs are typically produced using recombinant DNA technology in yeast (*Saccharomyces cerevisiae*) or baculovirus expression systems. The vaccine works by inducing high titers of type-specific neutralizing antibodies that prevent the virus from entering host cells, thereby preventing persistent infection and the development of cervical intraepithelial neoplasia (CIN). Developed and studied by companies including Merck and GSK, this monovalent formulation demonstrated high efficacy in landmark Phase II trials before its components were integrated into multivalent vaccines like Gardasil and Cervarix.
