Overview

This recombinant protein subunit vaccine component consists of virus-like particles (VLPs) of the Human Papillomavirus (HPV) type 18 L1 major capsid protein. The protein is produced using a baculovirus expression system in insect cells and self-assembles into VLPs that mimic the viral structure to induce a potent humoral immune response. It is formulated with the proprietary AS04 adjuvant system, which combines aluminum hydroxide with 3-O-desacyl-4'-monophosphoryl lipid A (MPL), a Toll-like receptor 4 (TLR4) agonist. MPL enhances the magnitude and duration of the antibody response by stimulating innate immunity and promoting a Th1-biased adaptive response. This specific entity is a primary constituent of the bivalent HPV vaccine Cervarix, developed by GSK, and is designed to prevent infections and associated malignancies, such as cervical cancer, caused by the high-risk HPV type 18. The Public Health Service of Amsterdam (Geneeskundige En Gezondheidsdienst Amsterdam) has been a significant investigator in clinical trials evaluating the efficacy and long-term immunogenicity of AS04-adjuvanted HPV vaccines.
