The human parainfluenza virus type 3 (HPIV3) Ebola vaccine is an experimental live-attenuated viral vector vaccine developed by the National Institute of Allergy and Infectious Diseases (NIAID). It is engineered using a recombinant HPIV3 backbone to express the glycoprotein (GP) of the Zaire ebolavirus. Unlike many other Ebola vaccines that are administered intramuscularly, this candidate is specifically designed for intranasal or aerosol administration to target the respiratory mucosa, aiming to elicit a robust local immune response in addition to systemic immunity. The HPIV3 vector is typically attenuated to prevent disease while remaining capable of limited replication to enhance immunogenicity. This vaccine platform is being evaluated for its potential to provide a needle-free alternative for Ebola prevention and to potentially block transmission through respiratory routes.
