Overview

A human prostate-specific membrane antigen plasmid DNA vaccine is a therapeutic cancer vaccine consisting of a plasmid encoding the full-length human prostate-specific membrane antigen (PSMA). Upon administration, the encoded PSMA protein is expressed in host cells, stimulating an immune response—primarily T-cell mediated—against cells expressing PSMA. This approach aims to induce both antibody and cytotoxic T-cell responses targeting prostate cancer cells that overexpress PSMA. The primary indication for this investigational therapy is recurrent or advanced prostate cancer. Clinical trials have demonstrated that such vaccines can elicit immunological responses and are generally well tolerated, with most side effects being mild and related to injection site reactions or flu-like symptoms[2][5][6].
