Human T lymphoid progenitors (HTLPs) are an investigational, ex vivo-generated allogeneic cell therapy designed to accelerate immune reconstitution in patients undergoing hematopoietic stem cell transplantation (HSCT) or suffering from severe combined immunodeficiency (SCID). Developed by researchers at Assistance Publique - Hôpitaux de Paris (AP-HP) and licensed to Smart Immune for development on their ProTcell platform, these cells are produced by culturing CD34+ hematopoietic stem and progenitor cells (derived from sources such as umbilical cord blood or mobilized peripheral blood) ex vivo for 7 days in a feeder-free system containing the Notch ligand Delta-like ligand 4 (DLL4-Fc) and a specific cytokine cocktail. Upon intravenous infusion, these early lymphoid progenitors migrate to the patient's thymus, where they undergo differentiation and selection to rapidly generate a diverse, self-tolerant, and functional polyclonal naive T-cell repertoire, potentially reducing the window of post-transplant immunodeficiency from over a year to approximately 100 days.
