Human umbilical cord-derived mesenchymal stem cells (hUC-MSCs) is an investigational cell therapy being developed by Beijing 302 Hospital (also known as The Fifth Medical Center of Chinese PLA General Hospital) for the treatment of liver failure, including acute-on-chronic liver failure (ACLF) and decompensated liver cirrhosis. The therapy involves the intravenous infusion of allogeneic MSCs derived from the Wharton's jelly of human umbilical cords. These cells are expanded in vitro in GMP-compliant facilities and administered in multiple infusions. The mechanism of action is multifaceted, primarily involving immunomodulation and tissue regeneration. The cells suppress systemic inflammation by inhibiting pro-inflammatory cytokines such as tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), and interleukin-17 (IL-17), and by modulating signaling pathways including JAK-STAT and PI3K-Akt. Clinical evaluations have demonstrated improvements in liver function and survival rates in patients with end-stage liver disease.
