Human umbilical cord mesenchymal stem cells (hUC-MSCs) are an allogeneic cell-based biologic therapy developed by Dr. Lingyun Sun and colleagues at the Affiliated Drum Tower Hospital of Nanjing University Medical School. These mesenchymal stem cells are isolated from human umbilical cord tissue and possess potent immunomodulatory properties. The therapy is primarily investigated for the treatment of refractory lupus nephritis (LN), particularly classes III, IV, and V. The mechanism of action involves the suppression of overactive immune responses by increasing T-regulatory (Treg) cell populations, reducing Th1 and Th17 cell subsets, and modulating B cell function and antibody production. Clinical studies, including Phase 2 randomized trials, have evaluated its efficacy and safety compared to standard immunosuppressants like mycophenolate mofetil (MMF), showing promising response rates in patients who are refractory to conventional therapy.
