Overview

Humanized anti-BCMA CAR-T cells (specifically the construct lenti-BCMA-2rd-CAR) is an autologous second-generation chimeric antigen receptor (CAR) T-cell therapy developed by Shanghai Genbase Biotechnology for the treatment of relapsed or refractory multiple myeloma (R/R MM). The therapy involves the genetic engineering of a patient's own T cells via lentiviral transduction to express a CAR targeting B-cell maturation antigen (BCMA), which is highly and specifically expressed on the surface of malignant plasma cells. By employing a humanized single-chain variable fragment (scFv) rather than a murine-derived one, the treatment is designed to reduce the risk of immunogenicity and the formation of anti-drug antibodies, potentially enhancing the persistence and long-term efficacy of the CAR-T cells. Clinical evaluations (such as trials ChiCTR1800017051 and ChiCTR2000033925) have demonstrated high overall response rates in R/R MM patients, although durability varies in patients with complex presentations such as extraosseous extramedullary disease.
