Humanized anti-CD19 CAR T cells are a type of genetically engineered T-cell therapy designed to treat relapsed/refractory acute lymphoblastic leukemia (r/r ALL). These T cells express a chimeric antigen receptor (CAR) that specifically targets the CD19 protein found on malignant B cells. The CAR incorporates a humanized single-chain variable fragment (scFv), derived from FMC63, which contains only 25% murine sequence, aiming to reduce immunogenicity and improve the persistence of the CAR T cells in patients. The CAR structure also includes CD8 hinge, CD8 transmembrane domain, CD137 costimulatory signal, and CD3 zeta intracellular domains to promote T-cell activation and sustained anti-tumor activity.
