Drug intelligence / Profile preview
humanized anti-VWF D4 monoclonal antibody (derived from the murine antibody mAb508) is a humanized monoclonal antibody designed to target the D4 domain of von Willebrand factor (VWF). Developed by INSERM, Université Paris-Saclay, and the University of Lille, it is being investigated for the prevention of shear-induced VWF degradation in patients undergoing mechanical circulatory support (MCS) or those with acquired von Willebrand syndrome (aVWS). Under high shear stress conditions, such as those induced by MCS devices, VWF undergoes excessive proteolysis by the metalloprotease ADAMTS13, leading to the loss of high-molecular-weight (HMW) VWF multimers and subsequent bleeding complications. By binding to the VWF D4 domain, this antibody partially blocks the VWF-ADAMTS13 interaction, preserving HMW VWF multimers and maintaining hemostatic function without completely abolishing physiological VWF degradation.
