Overview

Humanized CD19 CAR-T cells with TLR2 is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy developed by Zhujiang Hospital. The therapy consists of T cells genetically engineered to express a humanized CAR targeting the CD19 antigen, which is expressed on the surface of B-cell lineage cells. Uniquely, this CAR construct incorporates a Toll-like receptor 2 (TLR2) signaling domain alongside standard T-cell activation components. The inclusion of the TLR2 domain is intended to enhance the expansion, persistence, and cytotoxic activity of the CAR-T cells by providing potent costimulatory signals. It is currently being investigated in a Phase 1 clinical trial for the treatment of adult patients with newly diagnosed B-cell acute lymphoblastic leukemia (B-ALL) or B-cell lymphoblastic lymphoma (B-LBL) who are ineligible for or intolerant to intensive chemotherapy regimens.
