Overview

Humanized CT3 AbTCR-T cells are an experimental adoptive cell therapy targeting Glypican-2 (GPC2), a cell-surface protein frequently expressed in neuroblastoma and other pediatric solid tumors. The therapy utilizes the Antibody-T cell receptor (AbTCR) platform, which replaces the traditional single-chain variable fragment (scFv) used in CAR-T cells with a humanized antibody Fab fragment (CT3) fused to the constant regions of a γ/δ T-cell receptor. This construct also incorporates a CD30 co-stimulatory domain. By leveraging the physiological signaling machinery of the TCR complex rather than the non-physiological signaling of standard CARs, these cells demonstrate enhanced sensitivity to low antigen density, reduced exhaustion (lower expression of PD-1, LAG3, and TIM3), and superior persistence in solid tumor environments.
