Overview

HuMax-CD32b is a fully human monoclonal antibody developed by Genmab that targets CD32b (also known as FcγRIIb), the only inhibitory member of the Fcγ receptor family. CD32b is expressed on B cells and is often overexpressed in B-cell malignancies, where it functions to downregulate immune responses and can interfere with the efficacy of other therapeutic antibodies by internalizing them or providing inhibitory signals. HuMax-CD32b blocks these inhibitory pathways, thereby enhancing the anti-tumor activity of co-administered antibodies (such as those targeting CD20 or CD38) and potentially inducing direct apoptosis in malignant cells. Although it showed promise in preclinical models for various B-cell lymphomas and leukemias, the development of this specific candidate was eventually discontinued by Genmab.
