**HuMax-CD74** is a fully human monoclonal antibody targeting **CD74**, a chaperone protein for MHC class II and cell surface receptor for macrophage migration inhibitory factor (MIF), which is overexpressed on various hematological malignancies including B-cell lymphomas, leukemias, and multiple myeloma, as well as some solid tumors. Developed by **Genmab**, it binds CD74 to potentially inhibit MIF signaling and promote antibody-dependent cellular cytotoxicity (ADCC), though it has primarily been evaluated as the antibody component in antibody-drug conjugates (ADCs) such as those conjugated to auristatins or MMAF via linkers like MC-vc-PAB for enhanced cytotoxic delivery in cancers like non-Hodgkin lymphoma (NHL). Partnerships with Seattle Genetics and Novartis have explored ADC formats, but standalone clinical development status remains unclear with preclinical focus on hematologic cancers.[1][5][7][11]
