HuMax-ZP3 is a fully human monoclonal antibody developed by Genmab A/S that targets the Zona Pellucida glycoprotein 3 (ZP3). While ZP3 is naturally found in the extracellular matrix of oocytes, it is overexpressed in several solid tumors, including colon, pancreatic, and prostate cancers, but is absent in vital organs such as the brain, heart, and lungs. HuMax-ZP3 was engineered to trigger the immune system's killing mechanisms, specifically Antibody-Dependent Cellular Cytotoxicity (ADCC) and Complement Dependent Cytotoxicity (CDC), against ZP3-expressing tumor cells. Although it showed promising anti-tumor activity in pre-clinical SCID mouse models, the development of the drug was ultimately discontinued.
