Overview

huMIA-2 is a humanized monoclonal antibody that specifically targets the α3 domain of Major Histocompatibility Complex class I-related chain A and B (MICA/B). MICA and MICB are stress-induced ligands for the activating receptor NKG2D, which is expressed on Natural Killer (NK) cells and CD8+ T cells. In many cancers, tumor cells evade immune surveillance by proteolytically shedding the α1 and α2 domains of MICA/B from their surface. huMIA-2 binds to the α3 domain, which is proximal to the cell membrane, thereby preventing the proteolytic cleavage and subsequent shedding of the α1/α2 domains. By stabilizing MICA/B on the tumor cell surface, huMIA-2 restores and enhances NKG2D-mediated immune recognition and cytotoxic elimination of cancer cells. Preclinical studies have demonstrated its efficacy in reducing tumor growth and preventing metastasis in models of colorectal cancer, melanoma, and breast cancer.
