huMNC2-CD28-CD3z (also known as huMNC2-CAR28) is a preclinical-stage chimeric antigen receptor (CAR) T-cell therapy developed by Minerva Biotechnologies. It is designed to target MUC1* (mucin 1 star), a tumor-associated transmembrane cleavage product of MUC1 that acts as a potent growth factor receptor on solid tumors, particularly breast cancer. The construct incorporates a humanized MNC2 single-chain variable fragment (scFv) as the antigen-binding domain, a CD28 co-stimulatory domain, and a wild-type CD3-zeta (CD3z) signaling domain. In preclinical comparative studies, huMNC2-CD28-CD3z was evaluated alongside other constructs (such as the 4-1BB-containing huMNC2-CAR44 and the 1XX-mutated huMNC2-CAR22) to assess CAR-T cell exhaustion, persistence, and efficacy against low-antigen-expressing cancer cells.
