**HuNbTROP2-HSA-MMAE** is a preclinical TROP2-directed nanobody-drug conjugate developed for **TROP2-positive pancreatic cancer**. It comprises a humanized anti-TROP2 nanobody fused with an anti-human serum albumin nanobody to support systemic exposure, conjugated to the microtubule-disrupting payload monomethyl auristatin E through an MC-VC-PAB protease-cleavable linker with a drug-to-antibody ratio of 1. Following TROP2 binding and tumor-cell internalization, lysosomal processing releases MMAE, causing microtubule disruption and apoptosis associated with caspase-3 and caspase-9 activation. The agent showed potent antitumor activity in TROP2-positive pancreatic cancer cell models and xenografts, including complete tumor eradication at 5 mg/kg in a reported mouse xenograft study.
