Huwentoxin-IV (HWTX-IV) is a 35-amino acid peptide toxin originally isolated from the venom of the Chinese bird spider (*Cyriopagopus schmidti*, formerly *Haplopelma huwenum*). Developed by Shandong Sinobioway Biomedicine (a subsidiary of Beijing Sinobioway Group), the peptide is a potent and selective antagonist of the voltage-gated sodium channel Nav1.7 (SCN9A). Nav1.7 is a critical mediator of pain signaling in the peripheral nervous system, and its inhibition is a validated strategy for treating various pain conditions without the addictive potential of opioids. HWTX-IV acts by binding to the voltage-sensing domain II of the channel, preventing its activation. The drug has been advanced into clinical development in China for the treatment of moderate-to-severe pain.
