Overview

hVEGF26-104 is a research-stage therapeutic cancer vaccine developed by VU University Medical Center. It consists of a truncated, synthetic three-dimensional (3D)-structured peptide mimic covering amino acids 26–104 of the human VEGF165 isoform (79 amino acids total) with a cysteine-knot fold that mimics the bevacizumab binding site. Formulated with the adjuvant RFASE (Raffinose Fatty Acid Sulphate Ester) in an oil-in-water emulsion, the vaccine is designed to induce a polyclonal antibody response against endogenous vascular endothelial growth factor-A (VEGF-A/VEGF165) to inhibit tumor angiogenesis. Although hVEGF26-104 demonstrated neutralizing antibody induction in nonhuman primates, a Phase I clinical trial (NCT02237638) in patients with advanced solid malignancies showed that the vaccine was safe and well tolerated but failed to induce seroconversion against native hVEGF165 or provide clinical benefit.
