Drug intelligence / Profile preview
HVEM-Fc is a recombinant fusion protein consisting of the extracellular domain of Herpesvirus Entry Mediator (HVEM, also known as TNFRSF14) fused to the Fc region of immunoglobulin G (IgG). HVEM acts as a molecular switchboard in the immune system, integrating both stimulatory and inhibitory signals depending on its interacting ligands. HVEM-Fc functions as a B and T lymphocyte attenuator (BTLA) agonist and a decoy receptor that sequesters BTLA in trans, while also potentially blocking inflammatory LIGHT (TNFSF14) signaling. Preclinical research has investigated HVEM-Fc for its immunomodulatory properties in various disease models. In experimental glomerulonephritis, HVEM-Fc treatment has been shown to stimulate BTLA on classical dendritic cells (cDC1), promoting an anti-inflammatory phenotype, reducing crescent formation, and ameliorating proteinuria. Conversely, in oncology models such as non-small cell lung cancer (NSCLC), HVEM-Fc has demonstrated anti-tumor efficacy by augmenting and prolonging T-cell activity, particularly when combined with PD-1 inhibitors.
