HW221043 is an orally bioavailable small-molecule inhibitor of Kinesin Family Member 18A (KIF18A), developed by the Hubei Institute of Biopharmaceutical Industry. KIF18A is a kinesin motor protein that plays a critical role in regulating chromosome alignment and spindle dynamics during mitosis. Inhibition of KIF18A leads to mitotic arrest and apoptosis, particularly in cancer cells exhibiting chromosomal instability (CIN), while sparing normal cells. HW221043 is currently being evaluated in a Phase I first-in-human clinical trial in China to assess its safety, pharmacokinetics, and preliminary antitumor activity in patients with advanced, relapsed, or metastatic solid tumors who have failed standard therapies.
