HWAP-I is a recombinant humanized monoclonal antibody that targets Nerve Growth Factor (NGF), a key mediator in the development and maintenance of chronic pain states. Developed by Xiamen Amoytop Biotech (formerly known as Xiamen Beida Zhi Road Bioengineering), HWAP-I is specifically being evaluated for the management of moderate to severe cancer-related pain. The antibody works by binding to circulating NGF, preventing it from interacting with its high-affinity receptor, tropomyosin receptor kinase A (TrkA), and its low-affinity receptor, p75, on peripheral nociceptors. This inhibition prevents the NGF-induced sensitization of pain-sensing neurons and the subsequent upregulation of pain-related neurotransmitters and ion channels. As a non-opioid analgesic, HWAP-I represents a targeted approach to treating severe pain that is often refractory to conventional therapies like opioids or NSAIDs.
