HWH486 is a Bruton's tyrosine kinase (BTK) inhibitor being developed primarily for autoimmune diseases. It works by inhibiting BTK, an enzyme crucial in immune cell signaling. A Phase I trial in healthy Chinese adults showed favorable safety and pharmacokinetic profiles across doses from 10 to 800 mg. The drug demonstrated strong binding to BTK, peaking within 0.5-4.0 hours. Pharmacokinetic parameters showed linear Cmax increase and non-linear AUC increase with dose. It was well-tolerated. HWH486 is currently in Phase II clinical trials for chronic spontaneous urticaria, with a dose-finding study actively recruiting. Phase I trials for rheumatoid arthritis also occurred previously.
