HWK-016 is an antibody-drug conjugate (ADC) designed to target the membrane-bound portion of **MUC16**, a glycoprotein frequently overexpressed in ovarian and other female-origin malignancies. By focusing on the membrane-bound fraction, HWK-016 aims to avoid the “antigen sink” problem that hampered earlier MUC16-targeted therapies. Its mechanism of action involves delivery of a DNA topoisomerase I inhibitor payload to cancer cells expressing MUC16, leading to DNA damage and cell death. HWK-016 represents a next-generation ADC aimed at improving targeting precision and reducing off-target toxicity, addressing significant unmet needs in ovarian cancer. The drug was originally developed by WuXi Biologics and is further developed by Whitehawk Therapeutics, following in-licensing. As of July 2025, HWK-016 is in preclinical development, with IND submission planned for late 2025 and initial focus on ovarian cancer[1][3][4][5][7][9][10].
