HX008 + irinotecan is a combination therapy used as a second-line treatment for advanced or recurrent gastric or gastroesophageal junction adenocarcinoma. HX008 is a monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor, functioning as an immune checkpoint inhibitor to boost anti-tumor immune responses. Irinotecan is a small molecule chemotherapeutic agent and a topoisomerase I inhibitor, inducing cytotoxicity in cancer cells by inhibiting DNA replication and causing DNA strand breaks. The regimen consists of HX008 200 mg intravenously every 3 weeks and irinotecan 160 mg/m² intravenously every 2 weeks. This combination has demonstrated promising efficacy and a manageable safety profile in phase II clinical studies for patients refractory to first-line platinum and/or fluoropyrimidine therapy[1][5][7].
