HX009 + IN10018 is an investigational combination therapy for advanced solid tumors and hematological malignancies. HX009 is a first-in-class, rationally designed bispecific antibody that targets both programmed cell death protein 1 (PD-1) and CD47. By simultaneously inhibiting PD-1 on T lymphocytes and blocking CD47 on tumor cells, HX009 aims to enhance anti-tumor immune responses by promoting T-cell activation and macrophage-mediated phagocytosis of tumor cells. The molecule features weakened CD47 binding to reduce toxicity while maintaining efficacy in the tumor microenvironment[6][8]. IN10018 is a selective ATP-competitive small molecule inhibitor of focal adhesion kinase (FAK), which disrupts FAK signaling involved in cancer cell survival, metastasis, fibrosis, and immune evasion[7]. The combination is being evaluated in clinical trials for patients with advanced solid tumors—including biliary tract cancers—and relapsed/refractory lymphoma[2][3][4]. Early data suggest potential synergistic effects when combining these agents due to their complementary mechanisms.
