HX029 is an investigational tetra-specific antibody developed by Hanx Biopharmaceuticals (HanxBio). It is engineered to simultaneously target four distinct pathways involved in tumor progression and immune evasion: PD-L1, CTLA-4, VEGF, and CD28. By blocking the PD-L1 and CTLA-4 checkpoints, HX029 aims to restore T-cell mediated anti-tumor immunity. The inclusion of a VEGF-targeting domain provides anti-angiogenic activity, disrupting the tumor's blood supply and potentially normalizing the tumor microenvironment. Furthermore, the CD28-binding component is designed to provide costimulatory signals to T-cells, enhancing their activation and proliferation specifically within the tumor site. This multi-pronged approach is intended to overcome the limitations of monotherapy and provide a more robust therapeutic effect in patients with advanced solid tumors.
