HX116 is a novel multi-modal bispecific antibody-drug conjugate (BsAb-ADC) designed for the treatment of pan-solid tumors. Developed by Crown Bioscience, the molecule targets both Programmed Death-Ligand 1 (PD-L1) and Vascular Endothelial Growth Factor (VEGF), combining immune checkpoint inhibition and anti-angiogenesis with the targeted delivery of a cytotoxic payload. HX116 utilizes a bispecific antibody backbone conjugated to a moderate-potency topoisomerase I (TOPO-I) inhibitor with an optimized drug-to-antibody ratio (DAR). This multi-modal approach is intended to enhance drug internalization and anti-tumor efficacy compared to naked bispecific antibodies or individual monoclonal treatments. Preclinical data presented at the AACR 2026 meeting demonstrated robust cytotoxicity in 3D-tumor organoids and significant anti-tumor activity in humanized xenograft models, supporting its potential as a next-generation cancer therapy.
