Overview

HX1408 is an orally bioavailable, brain-penetrable small molecule inhibitor of poly [ADP-ribose] polymerase 1 (PARP1), developed by Fujian Haixi New Drug Creation. It is specifically engineered to cross the blood-brain barrier (BBB), addressing a critical limitation of many existing PARP inhibitors in the treatment of central nervous system (CNS) malignancies. By selectively inhibiting PARP1, HX1408 prevents the repair of single-strand DNA breaks, which subsequently progress to lethal double-strand breaks, inducing synthetic lethality in tumor cells with DNA damage response deficiencies. The drug is currently being evaluated in Phase I/II clinical trials in China for the treatment of recurrent or progressive glioblastoma in patients who have failed standard-of-care therapies, including surgery, radiotherapy, and temozolomide.
