Overview

HXB-319 is an investigational "trained" mesenchymal stromal cell (MSC) therapy developed by HexaBio for the treatment of systemic lupus erythematosus (SLE) and its severe complications, including lupus nephritis and diffuse alveolar hemorrhage (DAH). Unlike naïve MSCs, HXB-319 is engineered using a high-dimensional design of experiments (DoE) methodology to optimize the expression of therapeutic factors that mitigate inflammation and promote tissue repair. The therapy works by upregulating anti-inflammatory, angiogenic, and anti-fibrotic proteins such as tumor necrosis factor-inducible gene 6 protein (TSG-6), vascular endothelial growth factor (VEGF), and hepatocyte growth factor (HGF). Preclinical data indicates that HXB-319 modulates the immune system by reducing the populations of plasmacytoid dendritic cells and Th17 cells, increasing regulatory T cells (Tregs), and significantly lowering levels of pro-inflammatory mediators like IFN-gamma, IL-17A, and BAFF, thereby improving survival and preventing end-stage organ damage.
