Overview

HXN-1001 is a next-generation humanized monoclonal antibody targeting TL1A (tumor necrosis factor superfamily member 15, also known as VEGI). It is engineered for strong blocking activity, sub-nanomolar affinity for both trimeric and monomeric TL1A, and extended in vivo half-life. HXN-1001 demonstrates superior potency and efficacy compared to benchmark anti-TL1A antibodies and anti-IL-23 monoclonal antibodies in preclinical models of inflammatory bowel disease (IBD), ulcerative colitis, Crohn's disease, and psoriasis. The antibody functions by inhibiting TL1A-induced activation of the DR3-NFκB pathway, apoptosis in TF-1 cells, and IFN-γ secretion in human primary T cells. HXN-1001 is formulated at high protein concentration for subcutaneous administration. Developed using AI-guided protein engineering to optimize affinity, specificity, and pharmacokinetics, it is currently in phase 1 clinical trials as a therapeutic for several autoimmune and inflammatory diseases[1][3][4][5][6].
