HXN-1003 is a bispecific tetravalent antibody designed to simultaneously target and inhibit both interleukin 23 (IL23) and tumor necrosis factor superfamily member 15 (TL1A, also known as VEGI). The anti–IL23 arm specifically binds the p19 subunit of IL23 with high affinity, while the anti-TL1A arm binds both trimeric and monomeric TL1A with sub-nanomolar affinity. This dual targeting approach aims to enhance clinical efficacy in autoimmune diseases by overcoming resistance seen with single-target therapies. Preclinical studies have demonstrated that HXN-1003 provides synergistic or additive therapeutic effects in models of colitis and psoriasis, outperforming benchmark monoclonal antibodies such as risankizumab. The drug is being developed for autoimmune indications including colitis and psoriasis, particularly for patients refractory to existing treatments[2][3][5][7].
