HXN-1011 is a novel biparatopic antibody developed by Earendil Labs (affiliated with Helixon Therapeutics) that targets thymic stromal lymphopoietin (TSLP). Unlike conventional TSLP antibodies such as tezepelumab, which bind only to Site I, HXN-1011 was generated using AI algorithms to simultaneously bind to both Site I (the TSLPR binding site) and Site II (the IL-7RA binding site) of the TSLP cytokine. This dual-epitope targeting mechanism results in a binding affinity approximately 100 times higher than that of tezepelumab. Preclinical studies in mouse models of asthma have demonstrated that HXN-1011 effectively reduces lung inflammation, outperforming existing therapies like tezepelumab and GSK5784283. The drug is currently in preclinical development for the treatment of severe asthma and chronic obstructive pulmonary disease (COPD).
