HXN-1012 is a bispecific antibody developed by Earendil Labs (affiliated with Helixon Therapeutics) that simultaneously targets both thymic stromal lymphopoietin (TSLP) and interleukin-13 (IL-13) for the treatment of chronic respiratory diseases including asthma and chronic obstructive pulmonary disease (COPD). The drug is engineered as a "2+1" bispecific antibody capable of simultaneously binding to IL-13 and two non-overlapping epitopes on TSLP using an anti-TSLP biparatopic antibody and an anti-IL-13 antibody as building blocks. HXN-1012 is designed to inhibit both TSLP and IL-13 signaling pathways implicated in airway inflammation, with each arm exhibiting activity comparable to parent antibodies. In preclinical in vitro assays, HXN-1012 demonstrated superior activity compared to individual parent antibodies and showed more complete inhibition of inflammatory markers like TARC expression in PBMCs stimulated with TSLP and IL-13 compared to benchmark multispecific antibodies. The drug is currently in preclinical development and was developed using AI-guided engineering and optimization algorithms.
