HXN-1022 is a tetravalent bispecific antibody developed by Earendil Labs (an affiliate of Helixon) for the treatment of atopic dermatitis and other autoimmune skin disorders. Designed using an AI-guided platform, the antibody simultaneously targets and neutralizes OX40 ligand (OX40L) and Interleukin-31 (IL-31). By inhibiting OX40L, HXN-1022 aims to block T-cell co-stimulation and the underlying inflammatory response, while the neutralization of IL-31 specifically targets the itch-scratch cycle and pruritus associated with chronic skin inflammation. In preclinical studies, the drug has demonstrated high binding affinity for both targets and therapeutic efficacy in human PBMC-reconstituted Graft-versus-Host Disease (GvHD) mouse models, significantly inhibiting T-cell proliferation and prolonging survival.
