HXN-1031 is a trispecific antibody developed by Earendil Labs (a subsidiary of Helixon) currently in preclinical development for the treatment of B-cell related diseases. The molecule is engineered to simultaneously bind to two distinct B-cell surface antigens, CD19 and B-cell maturation antigen (BCMA), while also engaging the CD3 receptor on T cells. This trispecific configuration is designed to facilitate the formation of an immunological synapse between cytotoxic T cells and target B cells, leading to T-cell activation and the subsequent lysis of the pathogenic B cells. By targeting both CD19 and BCMA, HXN-1031 aims to enhance therapeutic efficacy and mitigate the risk of tumor escape through antigen loss, a common resistance mechanism in monospecific B-cell directed therapies.
