Overview

HXN-2009 is a biparatopic T-cell engager (TCE) being developed by Earendil Labs (a Helixon company) for the treatment of small-cell lung cancer (SCLC). The molecule is engineered to simultaneously bind to two distinct epitopes on Delta-like ligand 3 (DLL3), a Notch pathway ligand that is highly expressed in SCLC and other neuroendocrine tumors while remaining largely absent in healthy tissues. By engaging CD3 on the surface of T cells, HXN-2009 facilitates the formation of an immunological synapse between cytotoxic T cells and DLL3-positive tumor cells, leading to targeted tumor cell lysis. The biparatopic design is intended to improve binding affinity and selectivity compared to monoparatopic agents, potentially enhancing therapeutic efficacy in the preclinical setting.
