HXN-2010

HXN-2010 is a biparatopic antibody-drug conjugate (ADC) being developed by Earendil Labs (Helixon) for the treatment of small-cell lung cancer (SCLC). The drug targets Delta-like protein 3 (DLL3), a Notch ligand that is highly expressed on the surface of SCLC and other neuroendocrine tumor cells but minimally expressed in healthy tissues. HXN-2010 features a biparatopic antibody design, meaning it binds to two distinct, non-overlapping epitopes on the DLL3 protein, which is intended to promote superior receptor clustering, internalization, and lysosomal delivery compared to conventional monospecific antibodies. Additionally, it employs a dual-payload strategy, carrying two different cytotoxic drugs to enhance anti-tumor activity and mitigate the development of drug resistance. It is currently in the preclinical (PCC) stage of development.
