HY-2-15 is a novel small molecule radioligand developed for the detection and imaging of alpha-synuclein (α-syn) aggregates, which are pathological hallmarks of synucleinopathies. It specifically targets a binding site designated as Site 9 on α-synuclein fibrils. Research conducted at the University of Pennsylvania indicates that HY-2-15 exhibits a unique selectivity profile; in human post-mortem brain tissue autoradiography, it binds significantly to α-syn inclusions in Multiple System Atrophy (MSA) samples but shows minimal binding in Parkinson's disease (PD) or Dementia with Lewy Bodies (DLB) tissues. This specificity suggests that HY-2-15 could serve as a specialized diagnostic tool to differentiate MSA from other Parkinsonian disorders and to monitor disease progression in clinical settings.
