Overview

HY-oAd is an engineered oncolytic adenovirus (oAd) designed for the treatment of solid tumors, specifically non-muscle-invasive bladder cancer (NMIBC) and pancreatic cancer. It is a gene therapy vector that co-expresses three therapeutic proteins: interleukin-12 (IL-12), granulocyte-macrophage colony-stimulating factor (GM-CSF), and relaxin (RLX). The inclusion of relaxin is intended to degrade the dense extracellular matrix (ECM) characteristic of many tumors, thereby improving the spread of the virus and the infiltration of immune cells. IL-12 and GM-CSF are potent cytokines that work to activate the innate and adaptive immune systems, promoting a "cold" tumor microenvironment to become "hot." HY-oAd is often studied in combination with other therapies, such as the GSK-3β inhibitor elraglusib or PD-L1-targeted radioimmunotherapy, to achieve synergistic antitumor effects.
