HY034 is an investigational **allogeneic CAR-T cell therapy** designed to simultaneously target **CD19** on B cells and **B-cell maturation antigen (BCMA)** on long-lived plasma cells. Developed by **Huayao Jiexin**, the therapy is primarily being advanced for the treatment of autoimmune diseases, including **systemic lupus erythematosus (SLE)**. HY034 is manufactured using the **PrecisionGENE** platform, which utilizes non-viral, site-specific integration technology to insert the CAR into the **TRAC locus**, thereby eliminating TCRαβ expression to prevent graft-versus-host disease (GvHD). To enhance in vivo persistence and avoid rejection by host T cells and NK cells, the therapy co-expresses a human virus-derived protein that down-regulates certain MHC molecules. This 'off-the-shelf' approach aims to provide a significantly lower cost of goods and higher production yield compared to traditional autologous CAR-T therapies.
