Overview

HY1770 is an orally bioavailable, small molecule allosteric inhibitor of Tyrosine Kinase 2 (TYK2) developed by Suzhou Huyun New Drug Research and Development for the treatment of autoimmune and inflammatory diseases, specifically plaque psoriasis. By binding to the JH2 pseudokinase domain of TYK2 rather than the highly conserved JH1 catalytic domain, HY1770 achieves high selectivity over other Janus kinase (JAK) isoforms (JAK1, JAK2, and JAK3). This selective inhibition blocks the signaling of interleukin-23 (IL-23), interleukin-12 (IL-12), and Type I interferons, which are key drivers in the pathogenesis of psoriasis. The drug is currently being evaluated in Phase 1/2 clinical trials to assess its safety, tolerability, and efficacy in patients with moderate-to-severe plaque psoriasis.
