Overview

Hyal-GEMesys is a genetically engineered cell therapy developed by researchers at the National Institutes of Health (NIH). It consists of mesenchymal stromal cells (GEMesys) modified to express hyaluronidase (Hyal), an enzyme that degrades hyaluronic acid (HA). Hyaluronic acid is a key component of the tumor extracellular matrix (ECM) that contributes to high interstitial fluid pressure and physical barriers to therapy. By degrading HA, Hyal-GEMesys remodels the tumor microenvironment, reduces hypoxia, normalizes tumor vasculature, and enhances the infiltration and activation of T cells. Preclinical studies in murine models of osteosarcoma, rhabdomyosarcoma, and pancreatic cancer have shown that Hyal-GEMesys can reduce tumor volumes and synergize with chemotherapy by disrupting the protective stroma.
