Drug intelligence / Profile preview
Hyal2-ADC is a first-in-class antibody-drug conjugate (ADC) targeting hyaluronidase 2 (Hyal2), a membrane-bound enzyme involved in the degradation of tumor-associated hyaluronan (HA) in the extracellular matrix. Hyal2 is highly expressed by both epithelial tumor cells and immunosuppressive myeloid-derived suppressor cells (MDSCs) within the tumor microenvironment. Hyal2-ADC consists of an anti-Hyal2 monoclonal antibody conjugated to a cytotoxic PNU payload. By selectively depleting Hyal2-expressing tumor cells and MDSCs, Hyal2-ADC normalizes HA metabolism, alleviates tumor-associated immunosuppression, and promotes a robust T-cell-mediated anti-tumor immune response. It is being developed for the treatment of advanced solid tumors, including bladder, prostate, breast, and brain cancers, particularly in combination with immune checkpoint inhibitors like anti-PD-1 therapy.
