Hyaluronic acid-17β-estradiol conjugate is an investigational **polymer-drug conjugate** in which 17β-estradiol is covalently linked to sodium hyaluronate. The conjugate was developed as a brain-directed, prolonged-release estradiol delivery approach intended to cross the blood-brain barrier and mitigate systemic adverse effects associated with conventional estrogen supplementation. In ovariohysterectomized rat models of estrogen-deficiency-associated cognitive impairment, intravenous HA-E2 improved spatial learning and memory and increased markers of cholinergic innervation and hippocampal synaptic plasticity. It remains a preclinical research-stage therapeutic and is not an approved medicine.
