Overview

Hyaluronic acid hydrogels are widely used as drug delivery systems due to their biocompatibility, biodegradability, and ability to encapsulate various therapeutic agents—including small molecules (e.g., NSAIDs), biologics (e.g., antibiotics), and nanocrystals—for sustained or targeted release[1][3][4]. The mechanism of action is primarily physical and structural; the gel provides localized retention and controlled release at the site of administration while also interacting with cell surface receptors such as CD44 to modulate inflammation, tissue repair, hydration, and cellular migration[5][6]. Indications include intra-articular treatment for osteoarthritis or inflammatory arthritis[1][4], wound healing applications (as dressings or scaffolds)[8], tissue engineering/regeneration (cartilage repair), ophthalmic procedures, cosmetic/aesthetic uses (as fillers), and experimental pulmonary drug delivery[2][5]. The specific properties—such as swelling behavior and mucoadhesiveness—make these hydrogels suitable for both local injection into joints/tissues or topical use on wounds.
