Hybri is an experimental recombinant fusion protein designed for the treatment of systemic lupus erythematosus (SLE) and lupus nephritis. It features a dual-action mechanism intended to restore immune tolerance: a CTLA4 domain that binds to CD80 and CD86 on antigen-presenting cells to block T-cell costimulation via CD28, and a PD-L2 domain that acts as an agonist for the PD-1 receptor to enhance coinhibitory signaling. Developed by researchers at the University of Barcelona and IDIBAPS, Hybri has demonstrated efficacy in preclinical mouse models (NZBWF1 and MRL/lpr), where it reduced proteinuria, anti-dsDNA antibodies, and renal inflammation, suggesting potential as a maintenance therapy for autoimmune disorders.
