The hybrid dendritic cell vaccine is an investigational immunotherapy developed by the University of São Paulo General Hospital (HCFMUSP) for the treatment of glioblastoma and high-grade astrocytomas. Unlike traditional autologous dendritic cell vaccines, this approach utilizes allogeneic dendritic cells (DCs) derived from healthy donors which are fused with the patient's own (autologous) tumor cells to create hybridomas. This fusion process allows the professional antigen-presenting cells to present a comprehensive array of patient-specific tumor-associated antigens to the immune system. By using healthy donor DCs, the vaccine aims to bypass potential immune dysfunction or DC defects often found in cancer patients. The vaccine is administered via intradermal injection and is currently being evaluated in Phase III clinical trials as a maintenance therapy following standard surgery and chemoradiotherapy, both as a monotherapy and in combination with the PD-1 inhibitor pembrolizumab.
