**Hydrocodone polistirex** is an extended-release ion-exchange resin complex of hydrocodone, a semisynthetic opioid agonist, primarily used as a centrally acting antitussive and analgesic in combination products. It binds hydrocodone to a sulfonated styrene-divinylbenzene copolymer matrix, enabling controlled release via the Pennkinetic System, which prolongs suppression of the cough reflex by acting on brainstem respiratory centers. Typically formulated with chlorpheniramine polistirex for relief of cough and upper respiratory symptoms associated with allergies or colds, it exhibits peak plasma concentrations around 3.4 hours post-dose with a half-life of approximately 4 hours, distinguishing it from immediate-release hydrocodone formulations.[1][3][4]
